Watch our latest Labroots webinar on LocIn (pronounced “Loch-In”) where our Director of Molecular Biology and Cell Engineering, Dr Tim London presents this new innovative targeted integration system.
LocIn is an advanced targeted integration system created by RoukenBio designed for the efficient generation of cell lines by retargeting pre-defined genomic loci, known as "landing pads." These landing pads incorporate a fluorescent protein marker, enabling the easy identification of integrations at genomic sites that support high transgene expression.
The system leverages Recombinase Mediated Cassette Exchange (RMCE) to precisely replace the landing pad cassette with a gene of interest. This approach ensures the complete removal of the original cassette during retargeting while exclusively integrating the desired gene. By eliminating positional and context-dependent effects, LocIn enables accurate studies of phenotypic outcomes, delivering reliable genotype-to-phenotype correlations.
LocIn was developed with clear differentiators in mind which help drug developers and biotherapeutic innovators benefit from:

LocIn is a powerful tool transforming the traditional timelines of cell line generation making it less time consuming and more efficient. This has applications for testing target molecules across various therapeutic areas, like oncology, immuno-oncology, autoimmune diseases, allergy, and inflammatory conditions.
