Associate Scientific Director of Biophysics, Dr Sophie Moncrieff has a PhD in Molecular Biology, focusing on characterisation of a tumour suppressor E3 ubiquitin ligase. She has been working in immunology focused CROs since 2013, in a variety of laboratory-based and project management positions. She has a particular interest in antibody Fc effector function assays, as well as protein-protein binding assays using SPR (Biacore) technology.

Proving biosimilar comparability requires more than assumed Fc silencing. Even widely used ‘silent’ mutations can retain residual activity, making advanced binding and functional assays essential to accurately characterise Fc properties and meet regulatory expectations.

Surface Plasmon Resonance (SPR) plays a vital role across drug discovery and development, from early candidate screening to detailed product characterisation, including antigen and Fc‑receptor interactions. For biosimilar development, ensuring that SPR methods used for comparative binding analyses are rigorously qualified is essential for regulatory acceptance. This research poster outlines the qualification of multiple SPR assays designed to measure Fcγ receptor, FcRn, and antigen binding to a monoclonal antibody.

Anti-drug antibodies (ADAs) can derail a drug’s success before it ever reaches patients. In this study, we built a sensitive SPR-based assay to detect pre-existing ADAs in treatment-naïve serum helping drug developers make smarter, safer choices early on. From chip selection to control validation, here’s how we tackled the challenge.
